Aroid Establishment Medium

A pre-formulated medium designed for the establishment stage of aroid tissue culture.

Developed as a convenient starting formulation for establishing aroid explants in vitro, with MS nutrients, vitamins, sucrose, and BAP already combined in to one ready-to-use formulation.

Aroid Establishment Medium provides a pre-formulated nutritional and hormonal environment designed as a general starting point for establishing aroid plant material in culture.

Instead of measuring individual components such as basal salts, vitamins, sucrose and BAP yourself, everything is combined into a single powder formulation.

Designed for Aroids

The formulation is designed as a general starting medium for a range of commonly propagated aroids, including:

Monstera · Philodendron · Alocasia · Anthurium · Epipremnum

Plant response can vary between species, cultivars, explant types and culture conditions. This formulation should therefore be considered a general starting formulation, rather than a species-specific formula.

When should I use Establishment Medium?

Establishment Medium is intended for the initial establishment stage of your tissue culture workflow.

What You'll Need

To make sure you succeed with tissue culture propagation, you will need the following equipment and materials:

Gelling agent: Agar or other gelling agent such as Geltrite .

Sterile workspace

• Still Air Box (SAB) or Laminar Flow Hood
• 70% Isopropyl Alcohol (IPA) for surface disinfection

Vessels & sealing

• Culture jars, tubes, or Magenta boxes (pre-sterilized)
• Breathable caps or Parafilm for sealing

Plant material

• Healthy aroid explant (nodal segment, shoot tip, or leaf section)
• Surface sterilization agents: 70% ethanol, 10–20% bleach solution, sterile distilled water

Equipment

• Scalpel with sterile blades
• Forceps / tweezers
• Sterile gloves and face mask
• Pressure cooker or autoclave (for sterilizing tools and vessels)
• pH meter and pH adjustment solutions (KOH / HCl)

Preparation 

Recipe for 1 liter Aroid Medium:

1. Measure 34.5g of aroid media. (22.2g for rooting medium)
2. Add 800ml of distilled/deionized water, dissolve thoroughly
   
3. Adjust pH to 5.6-5.8, use pH buffer for higher/lower pH. (recommend having a pH meter)
4. Slowly add 6-8g of agar if a solid medium is desired, heat to dissolve.
5. Fill the remaining 200ml for a total volume of 1L medium.
6. When fully dissolved, pour in to culture jars and sterilize the prepared medium using an autoclave/pressure cooker.
7. Allow the medium to cool before introducing plant material.

This recipe is based on achieving 1 liter of finished media solution. If less/more is desired, make sure to adjust all ingredients accordingly.
